Cortisone


Mechanism of action:

Cortisone is a glucocorticoid. In the body, cortisone is reduced to cortisol by 11β-hydroxysteroid dehydrogenase type 1 (11β-HSD1) in the liver and adipose tissue. Cortisol can diffuse across the cell membrane and bind to the intracellular glucocorticoid receptor (GR) in the cytoplasm, then translocate into the nucleus to regulate the transcription of multiple genes. This suppresses the expression of pro-inflammatory genes such as COX-2, IL-1, IL-6, and TNF-α, while promoting the expression of anti-inflammatory genes such as Annexin A1.
