Vandetanib


Mechanism of action:

Vandetanib is a multi-target tyrosine kinase inhibitor. Most medullary thyroid cancers carry activating mutations in the RET proto-oncogene, and vandetanib inhibits RET kinase autophosphorylation, thereby blocking downstream signaling pathways such as MAPK and PI3K/Akt and suppressing tumor cell proliferation and survival. Vandetanib also inhibits VEGFR-2, reducing tumor angiogenesis and diminishing the blood supply to the tumor.
