Methotrexate

氨甲蝶呤

Mechanism of action:

Methotrexate 是一種二氫葉酸還原酶抑制劑（dihydrofolate reductase inhibitor）。Methotrexate 會競爭性抑制二氫葉酸還原酶（dihydrofolate reductase），阻斷二氫葉酸（dihydrofolic acid）轉換為四氫葉酸（tetrahydrofolic acid）。由於四氫葉酸是嘌呤與胸腺嘧啶（dTMP）合成的必要輔因子，此抑制會導致 DNA 與 RNA 合成受阻。
