Pentostatin

噴司他丁

Mechanism of action:

Pentostatin 是一種腺苷脫氨酶抑制劑(adenosine deaminase inhibitor)。腺苷脫氨酶(adenosine deaminase)能負責將去氧腺苷(deoxyadenosine)去胺,維持嘌呤代謝平衡。Pentostatin 抑制腺苷脫氨酶之後,會造成細胞的去氧腺苷累積,dATP 在細胞內大量增加。過量的 dATP 會抑制核糖核苷酸還原酶(ribonucleotide reductase)的活性,造成 dTTP、dCTP、dGTP 相對不足,而影響 DNA 的合成。
