特比萘芬
Mechanism of action:
Terbinafine 是一種烯丙基胺類(allylamine)衍生物。Terbinafine 可抑制黴菌細胞膜合成中的角鯊烯環氧化酶(squalene epoxidase),此酵素負責將角鯊烯(squalene)轉換為 2,3-環氧鯊烯(2,3-epoxysqualene),該步驟是麥角固醇(ergosterol)合成路徑中的早期步驟。Terbinafine 抑制了角鯊烯環氧化酶,會導致後面的麥角固醇減少,使黴菌細胞膜結構不完整;除此之外,角鯊烯因為無法轉化為 2,3-環氧鯊烯而不斷累積,引起毒性,最終使真菌生長受抑並可造成細胞損傷。
