Topotecan

Mechanism of action:
Topotecan 是一種拓撲異構酶 I 抑制劑（topoisomerase I inhibitor），屬於喜樹鹼（camptothecin）半合成衍生物。拓撲異構酶 I 的正常功能是在 DNA 複製或轉錄時，暫時切開單股 DNA 釋放扭轉張力，再將 DNA 接回。Topotecan 會與拓撲異構酶 I-DNA 複合體結合，穩定已切開但尚未重新接合的狀態。這些暫時性的單股斷裂在 DNA 複製叉通過時，由於另一股無法承受複製叉前進所造成的張力，硬生被扯段而產生雙股 DNA 斷裂，最終造成細胞凋亡。
