Cetuximab

西妥昔單抗

Mechanism of action:

Cetuximab 是一種嵌合型單株抗體，靶向表皮生長因子受體（EGFR，epidermal growth factor receptor）。EGFR 會活化 RAS-RAF-MEK-ERK（MAPK）路徑，促進細胞增殖；PI3K-AKT 路徑，抑制細胞凋亡；JAK-STAT 路徑，調控基因表現與血管新生。Cetuximab 能專一性地與 EGFR 結合，阻止 EGFR 與 EGF 或 TGF-α 等配體的結合，使 EGFR 無法活化，並可促使 EGFR 內吞與表現量下調；同時其抗體結構也可能引發抗體依賴性細胞毒性（ADCC）等免疫效應。
