Clomiphene

Mechanism of action:

Clomiphene 是一種選擇性雌激素受體調節劑（selective estrogen receptor modulator），主要作用位於下視丘-垂體-性腺軸（hypothalamic-pituitary-gonadal axis，HPG axis）。Clomiphene 能與下視丘的雌激素受體（estrogen receptor）結合，阻斷內源性雌激素的作用。雌激素遭阻斷後，下視丘會增加促性腺激素釋放激素（gonadotropin-releasing hormone）的分泌，進而促使腦下垂體（pituitary）分泌更多的濾泡刺激素（FSH） 與黃體成長激素（LH）。
