Dasatinib
達沙替尼
Mechanism of action:
Dasatinib 是一種第二代酪胺酸激酶抑制劑（second-generation tyrosine kinase inhibitor）。Dasatinib 主要作用於 BCR-ABL 融合蛋白，該蛋白是慢性骨髓性白血病（chronic myeloid leukemia，CML）因染色體易位產生的費城染色體（Philadelphia chromosome，t(9;22)(q34;q11)）上的融合基因。BCR-ABL 是一種持續活化的酪胺酸激酶，會過度刺激細胞生長、抑制凋亡，導致白血病。Dasatinib 可與 BCR-ABL 的 ATP 結合位點結合，阻止其活化下游訊號分子（如 STAT5、PI3K、MAPK），使癌細胞失去增殖訊號並誘導細胞凋亡。
