Decitabine

地西他濱
Mechanism of action:
Decitabine 是一種去氧胞苷(deoxycytidine)類似物。Decitabine 在細胞內會經過去氧胞苷激酶(deoxycytidine kinase)磷酸化成 decitabine monophosphate,之後再由其他激酶陸續磷酸化成 decitabine triphosphate。Decitabine triphosphate 有機會取代胞嘧啶(cytosine)被摻入 DNA 中。當 decitabine 被摻入 DNA 後,會使 DNA 甲基轉移酶(DNA methyltransferase,DNMT)形成共價複合物而被功能性抑制,抑制新生 DNA 的甲基化,促使異常沉默的腫瘤抑制基因重新表達。
