Denosumab

地舒單抗

Mechanism of action:

Denosumab 是一種單株抗體,專一性結合 RANKL(receptor activator of nuclear factor κ-B ligand)。正常生理中,成骨細胞(osteoblasts)分泌 RANKL,之後和破骨細胞(osteoclast)前驅細胞的 RANK 結合,促進破骨細胞成熟與活化。破骨細胞負責骨質吸收(bone resorption)。當 denosumab 結合 RANKL 後,使 RANK 無法被活化,便能抑制破骨細胞生成、分化與存活。由於減少骨質吸收,即提高骨密度與骨強度。
