Leflunomide

來氟米特
Mechanism of action:
Leflunomide 本身為前驅藥,在體內迅速轉化為其主要活性代謝物 teriflunomide(A77 1726)。Teriflunomide 會選擇性抑制二氫乳清酸脫氫酶(dihydroorotate dehydrogenase),此酵素位於粒線體內,是嘧啶核苷酸(pyrimidine nucleotide)新生合成途徑的其中一種酵素。當二氫乳清酸脫氫酶被 teriflunomide 抑制後,嘧啶核苷酸的新生合成量下降,導致 DNA 與 RNA 合成受限。
