Lomustine

洛莫司汀

Mechanism of action:

Lomustine 是一種具烷化反應性的化合物(alkylating agent),屬於亞硝基脲類(nitrosourea)衍生物。Lomustine 在體內自發性分解,產生具有高度反應性的烷化中間產物,這些活性分子可與 DNA 上的鳥糞嘌呤(guanine)鹼基結合,導致 DNA 鹼基修飾與結構異常。Lomustine 能讓兩股 DNA 之間(interstrand)或同一股內(intrastrand)造成交聯(cross-linking),DNA 因此無法正常解旋、複製或轉錄而誘發細胞死亡。
