Metformin

二甲雙胍

Mechanism of action:

Metformin 是一種雙胍類(biguanide)衍生物。Metformin 進入肝細胞後,抑制粒線體複合體 I(complex I)的活性,降低 ATP 生成,使 AMP/ATP 比值上升,進而活化腺苷單磷酸活化蛋白激酶(AMP-activated protein kinase)。腺苷單磷酸活化蛋白激酶被活化後,會抑制與葡萄糖新生相關的基因表達,使肝臟葡萄糖輸出下降。Metformin 也可促進骨骼肌與脂肪組織中葡萄糖轉運蛋白4(GLUT4)的表達與轉位,增加葡萄糖進入細胞的效率,降低胰島素阻抗。
