Naftifine

萘替芬

Mechanism of action:

Naftifine 是一種烯丙基胺類（allylamine）衍生物。Naftifine 可抑制黴菌細胞膜合成中的角鯊烯環氧化酶（squalene epoxidase），此酵素負責將角鯊烯（squalene）轉換為 2,3-環氧鯊烯（2,3-epoxysqualene），該步驟是麥角固醇（ergosterol）合成路徑中的早期步驟。Naftifine 抑制了角鯊烯環氧化酶，會導致後面的麥角固醇減少，使黴菌細胞膜結構不完整；除此之外，角鯊烯因為無法轉化為 2,3-環氧鯊烯而不斷累積，引起毒性，最終使真菌生長受抑並可造成細胞損傷。
