Tacrolimus

他克莫司

Mechanism of action:

Tacrolimus 是一種鈣調磷酸酶抑制劑(calcineurin inhibitor),屬於大環內酯類免疫抑制劑(macrolide immunosuppressant)。Tacrolimus 進入 T 細胞後,先與細胞內的FKBP12(FK506-binding protein 12)結合,形成 tacrolimus-FKBP12 複合體。該複合體會抑制鈣調磷酸酶(calcineurin)的活性。正常情況下,鈣調磷酸酶會去磷酸化(dephosphorylate)NFAT(nuclear factor of activated T cells),使其進入細胞核啟動基因轉錄。Calcineurin 被 tacrolimus 抑制後,NFAT 無法被去磷酸化,無法進入細胞核,使 IL-2、IL-4、IFN-γ、TNF-α 等細胞激素轉錄下降。
