Temozolomide

替莫唑胺

Mechanism of action:

Temozolomide 是一種具烷化反應性的化合物(alkylating agent),屬於咪唑四嗪類(imidazotetrazine)衍生物。Temozolomide 在生理 pH 值下會自發性水解,轉化為活性甲基化中間體 MTIC(monomethyl triazeno imidazole carboxamide)。MTIC 會將 甲基基團加到 DNA 鹼基上,主要作用位點包括 O⁶-guanine、N⁷-guanine、N³-adenine,反覆啟動 DNA 修復循環,最終導致 DNA 斷裂、細胞週期停滯、細胞凋亡。
