Tolcapone

托卡朋

Mechanism of action:

Tolcapone 是一種兒茶酚-O-甲基轉移酶抑制劑（catechol-O-methyltransferase inhibitor），常作為左旋多巴（Levodopa）的輔助。由於週邊的兒茶酚-O-甲基轉移酶會分解左旋多巴，降低其進入中樞神經系統的比例，而 tolcapone 可抑制週邊的兒茶酚-O-甲基轉移酶活性，協助更多左旋多巴進入中樞神經轉換為多巴胺。此外，tolcapone 亦能抑制中樞神經系統內的兒茶酚-O-甲基轉移酶，延長多巴胺在突觸間的作用時間，進而提高中樞多巴胺訊號的穩定性與可利用性。
