Trastuzumab

曲妥珠單抗

Mechanism of action:

Trastuzumab 是一種抗 HER2 單株抗體（anti-HER2 monoclonal antibody）。人類表皮生長因子受體 2（HER2，human epidermal growth factor receptor 2）屬於 ErbB 受體家族，其活化依賴與其他家族成員（HER1/EGFR、HER3、HER4）的二聚化。Trastuzumab 結合 HER2 受體後，可抑制 HER2 過度活化所引發的下游致癌訊號傳導，包括 PI3K/AKT、MAPK/ERK，使腫瘤細胞的增殖能力下降，並更容易進入凋亡。因為 trastuzumab 為 IgG1 抗體，可透過 Fc 區域引起抗體依賴性細胞媒介細胞毒性作用（antibody-dependent cell-mediated cytotoxicity）。
