Trifluridine

三氟胸苷
Mechanism of action:
Trifluridine 是一種嘧啶核苷(pyrimidine nucleoside)類似物。Trifluridine 進入細胞後,會被胸苷激酶(thymidine kinase)等酵素逐步磷酸化為有活性的單磷酸與三磷酸衍生物。Trifluridine 的單磷酸型可抑制胸苷酸合成酶(thymidylate synthase),阻斷 dTMP 的生成,造成 DNA 合成所需胸苷酸不足。Trifluridine 的三磷酸型可被直接嵌入正在合成的 DNA 中,造成DNA 結構不穩定,最後使細胞或病毒失去複製能力。
